I assume that your map is for DOOM2 original format and the new sound is in wav format. Let's say you want to replace the weapon sound DSSHOTGN. Open your pwad with Slade3. Drag the new sound file to the bottom of the list of lumps. Rename the sound name to DSSHOTGN, it is still in wav format. Rightclick on the sound name and select Audio --> Convert WAV to Doom Sound.
